A G inhibitor reduces intimal hyperplasia in aortocoronary saphenous vein grafts

چکیده
T Objective: Approximately 50% of aortocoronary saphenous vein grafts are occluded 10 years after coronary revascularization surgery. Intimal hyperplasia, a critical component in saphenous vein graft failure, is defined by vascular smooth muscle cell proliferation, which is mediated in part by subunits of heterotrimeric G proteins (G ) and downstream effectors such as mitogen-activated protein kinases. A peptide consisting of the carboxyl-terminus of the -adrenergic receptor kinase ( ARKct) binds G , thereby inhibiting G signaling. Utilizing a recombinant adenovirus containing the coding sequence for the ARKct peptide (Ad ARKct), this study investigates whether treatment of the vein graft with Ad ARKct reduces intimal hyperplasia in a large animal model of aortocoronary saphenous vein graft intimal hyperplasia.

Gene delivery to aortocoronary saphenous vein grafts in a large animal model of intimal hyperplasia.
OBJECTIVE More than 50% of aortocoronary saphenous vein grafts are occluded 10 years after surgery. Intimal hyperplasia is an initial, critical step in the progression toward occlusion. To date, no clinically relevant large animal models of aortocoronary saphenous vein graft intimal hyperplasia have been fully characterized. Gene therapy holds promise as a novel treatment for aortocoronary saph...
متن کاملAccelerated intimal hyperplasia in aortocoronary internal mammary vein grafts in minipigs
BACKGROUND More than 50% of aortocoronary saphenous vein grafts are occluded 10 years after surgery. Intimal hyperplasia is the initial critical step in the progression toward occlusion. Internal mammary veins, which are physiologically prone to less hydrostatic pressure, may undergo an accelerated progression to intimal hyperplasia and thus be suitable for investigation of the mechanisms of ao...
متن کاملUse of intravascular ultrasound for in vivo assessment of changes in intimal thickness of angiographically normal saphenous vein grafts one year after aortocoronary bypass surgery.
OBJECTIVE To use intravascular ultrasound in vivo to evaluate changes in the intimal thickness of angiographically normal saphenous vein grafts one year after implantation. DESIGN Fifteen saphenous vein grafts in 12 patients were examined one month and 12 months after aortocoronary bypass graft surgery with intravascular ultrasound using a 30 MHz transducer. None of the grafts examined showed...
متن کاملHistological survey of the saphenous vein before its use as autologous aortocoronary bypass graft.
A histological examination has been carried out on the saphenous veins used in 150 consecutive patients undergoing aortocoronary bypass. Morphological changes were observed frequently and consisted mainly of fibrosis of the intima and of the medial longitudinal muscular layer. Intimal fibrosis was rarely severe enough to narrow the lumen significantly. Statistical analysis disclosed that the fi...
متن کاملIntraoperative transfection of vein grafts with the NFkappaB decoy in a canine aortocoronary bypass model: a strategy to attenuate intimal hyperplasia.
BACKGROUND The nuclear transcriptional factor NFkappaB is reported to play an important role in the expression of genes for neutrophil and macrophage chemotactic factors, adhesion molecules, and cell cycle-regulating proteins. In aortocoronary bypass surgery, the saphenous vein often develops vein graft disease. Here, we investigated whether transfection of a cis element decoy oligodeoxynucleot...
